Professor Daekee Lee's research lab focuses on the molecular mechanisms underlying signal transduction in cancer and inflammatory diseases, with a particular emphasis on the EGFR/ERBB family of receptor tyrosine kinases, metallothionein regulation, and NADPH oxidase-mediated redox signaling. The lab investigates how dysregulation of these pathways contributes to tumorigenesis, therapy resistance, and tissue injury, using genetically engineered mouse models and molecular cell biology approaches. Key research directions include the role of ERBB3 in colorectal cancer progression, the regulation of oxidative stress through Nox1/NoxO1 signaling, and the function of growth factors like epiregulin in development and disease. The lab integrates in vivo models with biochemical and genomic analyses to uncover novel therapeutic targets in cancer and inflammatory disorders.

Perforating artery infarcts, whether single or occurring in addition to pial or border-zone infarcts, are lesion patterns specific for MCA disease. This suggests that local branch occlusion and coexisting distal embolization may be a common stroke mechanism in patients with MCA disease.
Epiregulin, an epidermal growth factor family member, acts as a local signal mediator and shows dual biological activity, stimulating the proliferation of fibroblasts, hepatocytes, smooth muscle cells, and keratinocytes while inhibiting the growth of several tumor-derived epithelial cell lines. The epiregulin gene (Ereg) is located on mouse chromosome 5 adjacent to three other epidermal growth factor family members, epigen, amphiregulin, and betacellulin. Gene targeting was used to insert a lacZ
Pharmacologic blockade of EGFR or the closely related receptor ERBB2 has modest efficacy against colorectal cancers in the clinic. Although the upregulation of ERBB3, a pseudo-kinase member of the EGFR/ERBB family, is known to contribute to EGFR inhibitor resistance in other cancers, its functions in normal and malignant intestinal epithelium have not been defined. We have shown here that the intestinal epithelium of mice with intestine-specific genetic ablation of Erbb3 exhibits no cytological
Mechanisms of regulation of mouse metallothionein (MT)-I gene expression in response to bacterial endotoxin-lipopolysaccharide (LPS) were examined. Northern blot analysis of hepatic MT-I mRNA in interleukin (IL)-6 or tumour necrosis factor (TNF)-receptor type I knock-out mice demonstrated that IL-6, not TNF-alpha, is of central importance in mediating hepatic MT-I gene expression in vivo after LPS injection. In vivo genomic footprinting of the MT-I promoter demonstrated a rapid increase, after L
The generation of reactive oxygen species (ROS) is required for proper cell signaling, but must be tightly regulated to minimize deleterious oxidizing effects. Activation of the NADPH oxidases (Nox) triggers ROS production and, thus, regulatory mechanisms exist to properly control Nox activity. In this study, we report a novel mechanism in which Nox1 activity is regulated through the proteasomal degradation of Nox organizer 1 (NoxO1). We found that through the interaction between NoxO1 and growt
ERBB3 is an emerging target for cancer therapy among the EGFR family. Contrary to resistance against EGFR and ERBB2 targeting, the genetic inhibition of ERBB3 results in anti-tumorigenic in HCT116 colon cancer cells harboring constitutively active KRAS and PIK3CA mutations. Still, the anti-tumorigenic molecular mechanism has not been defined. We demonstrated in this study that ERBB3 knockdown resulted in cell cycle arrest and activation of Bak and Bax-dependent apoptosis. Apoptosis was irrelevan
Isolated paresis of the inferior rectus may be caused by midbrain lesions such as metastasis1 or multiple sclerosis.2 Midbrain infarction is an extremely rare cause of isolated inferior rectus palsy.3,4 We report a patient with isolated inferior rectus palsy due to midbrain infarction probably involving the fascicle. The lesion was detected by diffusion-weighted MRI (DWI). A 56-year-old woman with hypertension and diabetes mellitus suddenly developed lightheadedness and diplopia on looking down
Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) causing coronavirus disease 2019 (COVID-19) has been a global health concern since 2019. The viral spike protein infects the host by binding to angiotensin-converting enzyme 2 (ACE2) expressed on the cell surface, which is then processed by type II transmembrane serine protease. However, ACE2 does not react to SARS-CoV-2 in inbred wild-type mice, which poses a challenge for preclinical research with animal models, necessitating a human
The role of metallothionein with regard to cadmium toxicity in vitro was investigated using preimplantation mouse blastocysts derived from a transgenic strain that constitutively overexpresses metallothionein-I transgenes (MT-I*). Northern blot and in situ hybridization revealed high levels of MT-I mRNA in transgenic blastocysts when compared with control blastocysts, and reverse transcriptase-polymerase chain reaction-amplified MT-I mRNA was almost exclusively MT-I*. Moreover, pulse-labeling ex
