Professor Ji Hyun Lee's research lab specializes in translational genomics and functional genomics, focusing on the genetic basis of cardiovascular and metabolic diseases, particularly familial hypercholesterolemia and lipid disorders in East Asian populations. The lab develops innovative CRISPR-based technologies—such as CRISPR/retron systems and CRISPR deaminase platforms—for high-throughput, multiplexed introduction and tracking of substitution mutations, enabling functional screening in human cell models. Their work integrates single-cell RNA sequencing, whole-exome sequencing, and systems biology approaches to uncover genotype-phenotype relationships and identify rare genetic variants underlying complex traits. The lab also investigates vascular phenotypes in autoimmune diseases, such as the link between rheumatoid arthritis and carotid atherosclerosis, using advanced imaging techniques.

Rare variants of APOB or PCSK9 were identified in nine of the 22 study patients with extremely low LDL-C levels, whereas most of them had common variants of the two genes. The common novelty of variants suggested polymorphism of the two genes among them. Our results provide rare genetic information associated with this lipid phenotype in East Asian people.
