Professor Sang Eun Yoon's research lab focuses on translational and clinical oncology, with a strong emphasis on understanding the molecular and immunological mechanisms underlying lymphomas, particularly T-follicular helper (Tfh)-derived lymphomas and primary central nervous system lymphoma (PCNSL). The lab investigates the role of the gut microbiome in modulating chemotherapy response and disease progression in diffuse large B-cell lymphoma (DLBCL), and explores biomarkers such as circulating tumor DNA (ctDNA) for improved monitoring and prognosis prediction. Additionally, the lab examines clinical predictors, including tumor laterality and molecular subtypes, to refine treatment strategies in colorectal and lymphoid malignancies.

Other TFH lymphomas appear to be a rare disease entity around one-quarter in nodal lymphomas of T follicular helper (Tfh) cell origin. Their less aggressive clinical feature than we did not expect is utterly different from PTCL-NOS and AITL. On the other hand, other TFH lymphomas share some characteristics, such as the cell of origin, a more common TBX21 subtype, and genetic variation such as RAS family mutation and epigenetic regulators, with AITL.
The gut microbiome influences cancer development and the efficacy and safety of chemotherapy but little is known about its effects on lymphoma. We obtained stool samples from treatment-naive, newly diagnosed patients with diffuse large B-cell lymphoma (DLBCL) (n = 189). The plasma ctDNA mutation analysis still has limited value for surveillance and predicting treatment outcomes of PCNSL because the detection efficiency was lower than other systemic lymphomas. Thus, analytical platforms should be improved to overcome anatomical hurdles associated with PCNSL.

The treatment outcome of patients with the Asian variant of IVLBCL is still not satisfactory. The prediction of CNS involvement based on the clinical features including CNS-IPI score might not serve to identify patients at high risk of CNS involvement, either.
