Professor Yutaka Satou's research lab focuses on the molecular and genomic mechanisms underlying chordate evolution and development, with a central emphasis on the ascidian Ciona intestinalis as a model organism. The lab investigates the genetic basis of key chordate innovations—such as notochord formation, heart specification, and neural development—by integrating genomics, functional analysis, and comparative biology. By leveraging high-quality genomic and transcriptomic resources, including cDNA clones, ESTs, and genome databases, the lab aims to decipher the regulatory networks governing early embryogenesis and the evolution of vertebrate complexity from simple chordate ancestors. The first chordates appear in the fossil record at the time of the Cambrian explosion, nearly 550 million years ago. Using these new resources, we identify a population of non-canonical (non-GT-AG) introns and also find that approximately 20% of Ciona genes reside in operons and that operons contain a high proportion of single-exon genes. An integrated genome database is essential for future studies of functional genomics. First, using morpholino against lacZ, we demonstrate that the morpholino specifically suppresses the translation of the gene introduced exogenously into Ciona eggs. Second, using morpholino against an alkaline phosphatase gene, we show that the morpholino suppresses the translation of the endogenous gene as well.
