Professor Dong-Seok Kim's research lab focuses on cellular signaling pathways and molecular mechanisms underlying skin pigmentation, neurodegenerative diseases, and cancer cell death. The lab investigates bioactive lipid mediators like sphingosine-1-phosphate and small molecules such as 4-n-butylresorcinol and novel compounds like KHG22394 to modulate melanogenesis through key signaling pathways including ERK and Akt. Sphingosine-1-phosphate (S1P) has emerged as a bioactive lipid modulator that mediates a variety of cell functions. However, the effects of S1P on melanogenesis are not well known. Therefore, we investigated the actions of S1P on melanin synthesis using a spontaneously immortalized mouse melanocyte cell line, Mel-Ab. This study shows that S1P significantly inhibits melanin synthesis in a concentration-dependent manner, and also that the activity of tyrosinase was reduced in S1P-treated cells. In

During our on-going attempts to develop a new skin-whitening agent, we identified a novel candidate compound KHG22394, a 2-imino-1,3-thiazoline derivative. Our data show that KHG22394 significantly inhibits melanin production in a dose-dependent manner, but that it does not directly inhibit tyrosinase, the rate limiting melanogenic enzyme.
