About the Lab

Professor Eung-seok Lee's research lab specializes in the development of novel therapeutic agents targeting cancer and inflammatory diseases. The lab focuses on designing and synthesizing small molecules with dual or multi-target inhibitory activities, particularly inhibitors of cyclooxygenase (COX) and lipoxygenase (LOX) for anti-inflammatory therapy, and topoisomerase IIα inhibitors for anticancer drug discovery. Their work emphasizes structure-activity relationship studies and the use of innovative synthetic methods, such as microwave-assisted synthesis, to develop potent, selective, and safer drug candidates. The lab also investigates molecular mechanisms of disease, including micrometastasis detection in gastric cancer and the regulation of inflammatory mediators like TNF-α and iNOS in macrophages.

BACKGROUND: Micrometastases consisting of one to a few cells in lymph nodes resected during gastrectomy are difficult to identify using conventional hematoxylin and eosin (H&E) stains. It has been shown that immunostaining for cytokeratins is effective in detecting lymph node micrometastasis in a variety of human tumors, but only a few previous reports demonstrated its use in the treatment of patients with early and advanced gastric carcinoma, and those reports had conflicting results. Lipopolysaccharide (LPS)-stimulated macrophages produce large amounts of nitric oxide (NO) by inducible nitric oxide synthase (iNOS). This is an important mechanism in macrophage-induced septic shock and inflammation. In the present study, we tested a synthetic propenone compound, 1-furan-2-yl-3-pyridin-2-yl-propenone (FPP-3) for its ability to inhibit the production of tumor necrosis factor-alpha (TNF-alpha) and an inducible enzyme, iNOS, in the LPS-stimulated murine macrophage-like cell line,
