About the Lab
Professor Gene Lee's research lab focuses on neuroendocrinology and molecular mechanisms underlying reproductive and endocrine physiology, with a particular emphasis on neuropeptides such as PACAP in ovarian function and steroidogenesis. The lab also investigates adrenal hormone regulation, especially aldosterone biosynthesis and its impact on development and systemic homeostasis, using genetically modified mouse models. Additionally, the lab explores stem cell biology in dental regeneration, aiming to harness pluripotent stem cells for epithelial cell differentiation. The integration of molecular endocrinology, developmental biology, and translational research defines the lab’s interdisciplinary approach.

15To study the effects of decreased amounts or absence of aldosterone on development and endocrine function, we have disrupted the mouse gene, Cyp11b2, coding for aldosterone synthase (AS) by replacing its first two exons with sequences coding for enhanced green fluorescent protein. The null pups fail to thrive postnatally, and about 30% die between d 7 and 28. Aldosterone in plasma and AS mRNA in adrenal glands are undetectable in the null mice. Adult AS-null mice are small, weigh 75% of wild typ
Tooth development and regeneration occur through reciprocal interactions between epithelial and ectodermal mesenchymal stem cells. However, the current studies on tooth development are limited, since epithelial stem cells are relatively difficult to obtain and maintain. Human embryonic stem cells (hESCs) and induced pluripotent stem cells (hiPSCs) may be alternative options for epithelial cell sources. To differentiate hESCs/hiPSCs into dental epithelial-like stem cells, this study investigated
Human Hertwig's epithelial root sheath/epithelial rests of Malassez (HERS/ERM) cells are epithelial remnants of teeth residing in the periodontium. Although the functional roles of HERS/ERM cells have yet to be elucidated, they are a unique epithelial cell population in adult teeth and are reported to have stem cell characteristics. Therefore, HERS/ERM cells might play a role as an epithelial component for the repair or regeneration of dental hard tissues; however, they are very rare population
Periodontal ligament stem cells (PDLSCs) are multipotent stem cells derived from periodontium and have mesenchymal stem cell (MSC)-like characteristics. Recently, the perivascular region was recognized as the developmental origin of MSCs, which suggests the <i>in vivo</i> angiogenic potential of PDLSCs. In this study, we investigated whether PDLSCs could be a potential source of perivascular cells, which could contribute to <i>in vivo</i> angiogenesis. PDLSCs exhibited typical MSC-like character
This paper summarizes the current state of the art in interplanetary supply chain modeling and discusses SpaceNet as one particular method and tool to address space logistics modeling and simulation challenges. Fundamental upgrades to the interplanetary supply chain framework such as process groups, nested elements, and cargo sharing, enabled SpaceNet to model an integrated set of missions as a campaign. The capabilities and uses of SpaceNet are demonstrated by a step-by-step modeling and simula
Human oral squamous cell carcinoma cell lines (KOSCC-11, -25A, -25B, -25C, -25D, -25E, -33A, and -33B) were established by explantation culture from these oral squamous cell carcinomas. The histopathology of the primary tumors, in vitro growth characteristics, epithelial origin, in vitro anchorage-independency, in vivo tumorigenicity, the frequency of human papillomavirus (HPV) infections, and the status of proto-oncogenes, tumor suppressor genes, DNA mismatch repair genes, and microsatellite in
Once tooth development is complete, odontoblasts and their progenitor cells in the dental pulp play a major role in protecting tooth vitality from external stresses. Hence, understanding the homeostasis of the mature pulp populations is just as crucial as understanding that of the young, developing ones for managing age-related dentinal damage. Here, it is shown that loss of Cpne7 accelerates cellular senescence in odontoblasts due to oxidative stress and DNA damage accumulation. Thus, in Cpne7-

Models and simulations are often classified by the US Department of Defense into four levels—campaign, mission, engagement, and engineering. In integrated modeling and simulation of space exploration architectures and campaigns, the challenge is to effectively explore the breadth of the trade space while reaching sufficient depth to gain insight. The use of Design of Experiments (DOE) in this field can enable more efficient parametric studies thereby allowing more resources to be focused on detailed analysis of interesting cases. This paper presents an approach based on DOE with orthogonal arrays and presents a recent integrated mode
Research on tooth regeneration using human-induced pluripotent stem cells (hiPSCs) is valuable for autologous dental regeneration. Acquiring mesenchymal and epithelial cells as a resource for dental regeneration is necessary because mesenchymal-epithelial interactions play an essential role in dental development. We reported the establishment of hiPSCs-derived dental epithelial-like cell (EPI-iPSCs), but hiPSCs-derived dental mesenchymal stem cells (MSCs) have not yet been reported. This study w
