About the Lab

Professor Seung-Jae V. Lee's research lab focuses on molecular mechanisms underlying aging and longevity, with a particular emphasis on the regulatory roles of non-coding RNAs, especially tRNA halves (tRHs). The lab investigates ribonucleases such as DIS-3/DIS3 in tRH generation and their downstream effects on translation regulation and stress response pathways like SKN-1/NRF. Additionally, the lab applies advanced computational and experimental methods to optimize water quality assessment and disinfection processes in water treatment systems, integrating fluid dynamics and sensory science. Their interdisciplinary work bridges molecular biology, environmental engineering, and systems biology to address both biomedical and environmental challenges.

Brief guide to RT-qPCR

RNA quantification is crucial for understanding gene expression and regulation. Reverse transcription-quantitative polymerase chain reaction (RT-qPCR) is a widely used technique for RNA quantification because of its practical and quantitative nature, sensitivity, and specificity. Here, we provide an overview of RT-qPCR, focusing on essential reagents, the importance of primer design, the detailed workflow, and data analysis methods. This guide will be useful for scientists who are unfamiliar wit

This study was motivated by the purpose of improving the O-Index, currently used to quantify water tasty. The authors first develop the M-Index after normalizing Ca, K, and SiO2 concentrations in the spring water, in that the concentrations are subject to their log-normal distributions. The M-Index is then compared with the O-Index based on the results of sensory tests, revealing that sensory tests are correlated with the M-Index much more than the O-Index. Furthermore, the developed index is ap

Disinfection is a basic and effective microorganism inactivation method and historically contributed a decrease in waterborne diseases. To guarantee the disinfection ability, improving T in CT value is important. Many indexes are used to estimate the hydraulic efficiency, however, these are black-box analysis. Therefore it is need to develope new estimation method. In this study, internal short-circuiting estimation method (ISEM) is developed using CFD and we inquire into the factor which causes

A large majority of clearwell must be modified with maintaining the present baffles since they were underground built with the material like concrete. Therefore it is unreasonable to apply the previous research in clearwell modification which is studied with the assumption that distance between baffles is constant. In this study, internal short-circuiting estimation method (ISEM), which has the advantage of being applied at any condition, is applied to evaluate modification of A clearwell and mo

Transfer RNA (tRNA) halves (tRHs) are generated via the cleavage of tRNAs, but their roles in aging and longevity remain poorly understood. Here, we demonstrate a direct role of tRHs in aging in metazoans. Through a genetic screen using Caenorhabditis elegans, we identify DIS-3/DIS3 as a ribonuclease that catalyzes tRH generation, including 5′-tRH-Gln and 5′-tRH-Asp, from tRNAs. Among them, 5′-tRH-Gln is essential for longevity conferred by various interventions, including dietary restriction. G
