研究室紹介
Professor Hyunsook Kim's research lab focuses on metabolic regulation, particularly the interplay between growth hormone, insulin-like growth factor-I (IGF-I), and lipid metabolism in the context of type 2 diabetes and obesity-related disorders. The lab investigates the therapeutic potential of natural compounds—such as pterostilbene, blueberry byproducts, and kefir-derived exopolysaccharides—in improving metabolic health, reducing adiposity, and modulating cholesterol and bile acid metabolism. Additionally, the lab explores the biological and behavioral impacts of these bioactive compounds using animal models and molecular metabolic analyses.

15Abnormalities in insulin action are the characteristics of type 2 diabetes. Dominant-negative muscle-specific IGF-I receptor (MKR) mice exhibit elevated lipid levels at an early age and eventually develop type 2 diabetes. To evaluate the role of elevated lipids in the progression of the diabetic state, MKR mice were treated with WY14,643, a peroxisome proliferator-activated receptor (PPAR)-alpha agonist. WY14,643 treatment markedly reduced serum fatty acid and triglyceride levels within a few da
Pterostilbene, 3',5'-dimethoxy-4-hydroxystilbene, is a resveratrol analogue and has been reported to have similar and often potent health-promoting properties. Pterostilbene has been shown to reduce weight gain, liver fat, plasma cholesterol, adiposity, inflammatory biomarkers, blood glucose, and other physiological characteristics of metabolic diseases in animal models. Studies on pterostilbene suggest that it may improve risk factors associated with diabetes, cardiovascular disease, fatty live
GH and IGF-I are potent regulators of muscle growth and function. Although IGF-I is known to mediate many of the effects of GH, it is not yet clear whether all effects of GH are completely dependent on the IGF-I system. To evaluate the biological effects of the GH/IGF-I axis on muscle growth, we administrated recombinant human GH to mice, which lack IGF-I function specifically in skeletal muscle, due to the overexpression of a dominant-negative IGF-I receptor in this tissue (MKR mice). GH treatm
Physiological properties of water-soluble exopolysaccharides (EPS) and residues after EPS removal (Res) from the probiotic kefir were determined in high-fat (HF) diet-fed C57BL/6J mice. EPS solutions showed rheological properties and lower viscosity compared to those of β-glucan (BG). EPS significantly suppressed the adipogenesis of 3T3-L1 preadipocytes in a dose-dependent manner. Mice were fed HF diets containing 5% EPS, 5% BG, 8% Res, or 5% microcrystalline cellulose (control) for 4 weeks. Com
This study analyzed plasma lipid profiles, genes related to cholesterol and bile acid metabolism, and inflammation in liver as well as adipose tissue from Syrian Golden hamsters fed high-fat diets supplemented with blueberry (BB) pomace byproducts including 8% dried whole blueberry peels (BBPWHL), 2% dried extract of peels (BBPX; 95% ethanol extract), and 6% residue from extracted peel (BBPEXT) compared to a diet containing 5% (w/w) microcrystalline cellulose (control). All BB diets significantl

The relationship between the intestinal microbiota and the hypocholesterolemic and antiobesity effects of whole grape seed flour from white and red winemaking was evaluated. Male Golden Syrian hamsters were fed a high-fat (HF) control diet or a HF diet supplemented with 10% partially defatted grape seed flours from either Chardonnay (ChrSd) or Cabernet Sauvignon (CabSd) grapes for 3 weeks. The numbers of total bacteria and relative abundances of Bifidobacterium spp., Lactobacillus spp., and Firm
The antiobesity and antidiabetic effects of the beta3-adrenergic agonists were investigated on nonobese type 2 diabetic MKR mice after injection with a beta3-adrenergic agonist, CL-316243. An intact response to acute CL-316243 treatment was observed in MKR mice. Chronic intraperitoneal CL-316243 treatment of MKR mice reduced blood glucose and serum insulin levels. Hyperinsulinemic euglycemic clamps exhibited improvement of the whole body insulin sensitivity and glucose homeostasis concurrently w
The mechanisms for the hypocholesterolemic and antiobesity effects of grape seed flours derived from white and red winemaking processing were investigated using male Golden Syrian hamsters fed high-fat (HF) diets supplemented with 10% partially defatted grape seed flours from Chardonnay (ChrSd), Cabernet Sauvignon (CabSd), or Syrah (SyrSd) pomace as compared to a HF control diet for 3 weeks. Hamsters fed the ChrSd diet had significantly lowered plasma total-, VLDL-, and LDL-cholesterol concentra
The antiobesity action of nonviable probiotic lactic acid bacteria (PLAB) may be attributed to bacterial cellular components recognized by host cells. The anti-inflammation and antiobesity properties of surface layer proteins (SLPs) that are cellular components isolated from kefir PLAB were determined in macrophage RAW 264.7 cells and obese mice. Kefir SLPs significantly decreased secretion of IL-6 and production of NF-kB p65 protein by LPS-stimulated RAW 264.7 cells in a dose-response manner. C
