Professor Jae-Kyung Won's research lab specializes in cancer genomics and molecular oncology, focusing on identifying novel genetic and molecular drivers in thyroid and lung cancers through integrated whole-genome and transcriptome sequencing. The lab investigates the mutational landscapes, RNA editing events, and gene expression alterations that contribute to tumorigenesis, disease progression, and therapeutic resistance in aggressive thyroid cancers and non-small cell lung cancer. A key emphasis is on translating genomic findings into clinically relevant biomarkers and therapeutic targets, particularly in cancers with limited known drivers or poor outcomes. The lab also explores the role of non-coding mutations, such as in the TERT promoter, and post-transcriptional modifications like RNA editing in cancer pathogenesis.

Follicular thyroid carcinoma (FTC) and benign follicular adenoma (FA) are indistinguishable by preoperative diagnosis due to their similar histological features. Abstract Anaplastic thyroid cancer (ATC) and advanced differentiated thyroid cancers (DTCs) show fatal outcomes, unlike DTCs. Here, we demonstrate mutational landscape of 27 ATCs and 86 advanced DTCs by massively-parallel DNA sequencing, and transcriptome of 13 ATCs and 12 advanced DTCs were profiled by RNA sequencing. The diagnostic performance of shear-wave and strain elastography was similar. Either elastography technique can improve overall diagnostic performance in the differentiation of benign and malignant lesions when combined with B-mode ultrasound. However, the sensitivity and specificity of shear-wave and strain elastography were different according to lesion histologic profile, tumor grade, and breast thickness.
BACKGROUND: Recent reports suggest that mutations in the promoter of the gene encoding telomerase reverse transcriptase (TERT) affect thyroid cancer outcomes. METHODS: In all, 551 patients with differentiated thyroid cancer (DTC) enrolled in this study. The median follow-up duration was 4.8 years (interquartile range, 3.4-10.6 years).
