研究室紹介

Professor Kichul Shin's research lab focuses on the immunological roles of mast cells in chronic inflammatory diseases, particularly rheumatoid arthritis and parasitic infections. The lab investigates mast cell heterogeneity, protease-mediated inflammation (especially tryptase and mMCP-6), and the therapeutic potential of mesenchymal stem cells in autoimmune arthritis. Key research directions include understanding the pathogenic mechanisms of mast cells in joint inflammation and evaluating stem cell-based therapies in clinical and preclinical models.

Mast Cells Contribute to Autoimmune Inflammatory Arthritis via Their Tryptase/Heparin Complexes

SJR Q1The Journal of ImmunologyOA

Although mast cells (MCs) often are abundant in the synovial tissues of patients with rheumatoid arthritis, the contribution of MCs to joint inflammation and cartilage loss remains poorly understood. MC-restricted tryptase/heparin complexes have proinflammatory activity, and significant amounts of human tryptase beta (hTryptase-beta) are present in rheumatoid arthritis synovial fluid. Mouse MC protease-6 (mMCP-6) is the ortholog of hTryptase-beta, and this serine protease is abundant in the syno

Mouse Mast Cell Tryptase mMCP-6 Is a Critical Link between Adaptive and Innate Immunity in the Chronic Phase of Trichinella spiralis Infection

SJR Q1The Journal of ImmunologyOA

Although the innate immune function of mast cells in the acute phase of parasitic and bacterial infections is well established, their participation in chronic immune responses to indolent infection remains incompletely understood. In parasitic infection with Trichinella spiralis, the immune response incorporates both lymphocyte and mast cell-dependent effector functions for pathogen eradication. Among the mechanistic insights still unresolved in the reaction to T. spiralis are the means by which

Intravenous Infusion of Umbilical Cord Blood-Derived Mesenchymal Stem Cells in Rheumatoid Arthritis: A Phase Ia Clinical Trial

SJR Q1Stem Cells Translational MedicineOA

Abstract Based on immunomodulatory actions of human umbilical cord blood-derived mesenchymal stem cells (hUCB-MSCs), in vitro or preclinical studies of hUCB-MSCs have been conducted extensively in rheumatoid arthritis (RA). However, few human trials have investigated the outcomes of hUCB-MSC infusions. The CURE-iv trial was a phase I, uncontrolled, open label trial for RA patients with moderate disease activity despite treatment with methotrexate. The patients received a single intravenous infus

Lymphocyte‐independent connective tissue mast cells populate murine synovium
Kichul Shin, Michael F. Gurish, Daniel S. Friend, Alan D. Pemberton, Elisabeth M. Thornton, H. R. P. Miller, David Lee
Arthritis & Rheumatism

OBJECTIVE: Mast cells (MCs) are a heterogeneous population of tissue-resident bone marrow-derived cells; distinct MC subpopulations are situated at specific microanatomic locations. The phenotype of the murine synovial MC remains undefined. Since MCs have been implicated in the pathogenesis of inflammatory arthritis, we sought to define the phenotype of the murine synovial MC population in normal and arthritic joints. We also examined the contribution of lymphocytes to synovial MC physiology. ME

SJR Q2BMC Musculoskeletal DisordersOA

Bone SPECT/CT is more useful than conventional bone scintigraphy in identifying sacroiliitis in early axial SpA patients, even with mild SI joint changes in plain radiography. By combining CT, we can accurately delineate the sacrum and SI joint uptake with our VOI method.

This study aimed to investigate drug retention rates for various TNF inhibitors (TNFis) commonly prescribed to Korean patients with ankylosing spondylitis (AS) in the Korean College of Rheumatology Biologics registry (KOBIO; December 2012–June 2016). Discontinuation was defined as switching or stopping the biologic agent. Kaplan–Meier curves and Cox's proportional hazard models were used for further analysis. The reasons for discontinuation of TNFis were also assessed. Univariate and multivariat

SJR Q3International Journal of Rheumatic Diseases

AIM: To investigate the efficacy and safety of tacrolimus (TAC) versus leflunomide (LEF) when combined with methotrexate (MTX) in rheumatoid arthritis (RA) patients. METHOD: This was a 24-week multi-center, double-blind, randomized, non-inferiority study targeting RA patients with moderate to severe Disease Activity Score of 28 joints (DAS28 > 3.2) who showed inadequate response to MTX. Patients were randomized into TAC or LEF (add-on to MTX) groups. Initial daily doses of TAC and LEF were 1.5 a

SJR Q2Planta Medica

A new method for separation and quantitative determination of flavone-C-glycosides in seeds of Zizyphus vulgaris var. spinosus by high performance liquid chromatography was elaborated. A reverse-phase system with a COPELL-ODS column using tetrahydrofuran in water (4:53) as a mobile phase was developed. The flavonoids were detected at 330 nm and the analysis was successfully carried out within 15 min.
