Professor Yang-Keun Kim's research lab focuses on viral pathogenesis, structural virology, and innate immune evasion, with a central emphasis on understanding the molecular mechanisms of viral proteins that interact with non-canonical nucleic acid structures such as Z-DNA and pseudoknots. The lab investigates how viral proteins like E3L from poxviruses exploit Z-DNA binding and dsRNA recognition to counteract host antiviral responses, particularly interferon signaling, and how these interactions influence viral pathogenicity. Additionally, the lab explores the structural and functional basis of ribosomal frameshifting in viruses and develops advanced microencapsulation technologies for microbial protection and biotechnological applications. The N-terminal domain of the E3L protein of vaccinia virus has sequence similarity to a family of Z-DNA binding proteins of defined three-dimensional structure and it is necessary for pathogenicity in mice. Zinc-finger proteins of the Cys2His2 type bind DNA-RNA hybrids with affinities comparable to those for DNA duplexes.
