김범준 교수의 연구실은 주로 결핵 및 비결핵성 미세박테리아의 분자생물학적 진단과 유전적 특성 분석을 중심으로 하며, rpoB 및 hsp65 유전자를 활용한 유전자 기반 종 구분 기법을 개발·응용하고 있습니다. 또한 한국에서 흔한 간염 바이러스 B의 유전자 변이 및 바이러스 유전자 변형이 간질환 진행에 미치는 영향을 분석하여, 임상적 결과와의 연관성을 규명하고 있습니다. 특히, HBV 유전자의 주요 항원 영역에서의 자연발생적 변이가 만성 간염, 간경변, 간세포암 등의 병태생리에 미치는 영향을 직접 시퀀싱 기반으로 연구하고 있습니다.

For the differentiation and identification of mycobacterial species, the rpoB gene, encoding the beta subunit of RNA polymerase, was investigated. rpoB DNAs (342 bp) were amplified from 44 reference strains of mycobacteria and clinical isolates (107 strains) by PCR. The nucleotide sequences (604 bp) of partial heat-shock protein genes (hsp65) from 161 Mycobacterium strains containing 56 reference Mycobacterium species and 105 clinical isolates were determined and compared. hsp65 sequence analysis showed a higher degree of divergence between Mycobacterium species than did 16S rRNA gene analysis. The RNA polymerase beta-subunit genes (rpoB) of 67 Streptomyces strains, representing 57 species, five Kitasatospora strains and Micromonospora echinospora KCTC 9549 were partially sequenced using a pair of rpoB PCR primers. Among the streptomycetes, 99.7-100 % similarity within the same species and 90.2-99.3 % similarity at the interspecific level were observed by analysis of the determined rpoB sequences.
