Byung Yoon Choi 교수의 연구실은 청각 장애의 유전적 기반을 규명하고, 특히 SLC26A4 및 GJB2 유전자 변이가 유발하는 청력 손실의 분자 기전과 임상적 연관성을 중심으로 연구를 진행하고 있습니다. 특히 비정형적 청력 손실과 관련된 유전자 변이의 유병률 및 병태생리학적 역할을 전반적으로 분석하며, 청각 보조 기기(청각 보청기, 인공와우)의 적정 시기 결정을 위한 유전자 기반 예측 모델 개발에도 기여하고 있습니다. 또한, 산전 유전자 진단 기술을 응용한 비침습적 선별 진단 방법의 개선을 통해 유전성 청각 장애의 조기 진단과 조기 간 interventions 가능성을 모색하고 있습니다.

Hearing loss with enlargement of the vestibular aqueduct (EVA) can be associated with mutations of the SLC26A4 gene encoding pendrin, a transmembrane Cl(-)/I(-)/HCO(3)(-) exchanger. Current molecular genetic testing including deafness panel sequencing helps to predict the 2-year follow-up outcomes after CI in prelingually deafened children. GD cochlear implantees show better functional outcomes after CI than undiagnosed cochlear implantees as determined by deafness panel sequencing, suggesting a genotype-functional outcome correlation. Most X-linked nonsyndromic hearing loss is caused by various types of mutations of the POU domain class 3 transcription factor 4 gene (POU3F4). We found five unique missense and frameshift truncation and extension mutations in Korean patients. Two missense mutations (p.Thr211Met and p.Gln229Arg) disturbed transcriptional activity. Two frameshift extension mutations (p.Thr354GlnfsX115 and p.X362ArgextX113) were located outside of POU domain and nuclear localization signal (NLS) at the C-terminus.
Our study further defines the spectrum of SLC26A4 mutations among East Asians and demonstrates a rapid and efficient protocol for their detection.
In the present study, we report that MYH14 can manifest as nonsyndromic prelingual severe sensorineural hearing loss in an AD fashion in Koreans. The results of the present study suggest that further genetic studies of similar patients should consider MYH14 as a causative gene, and cochlear implantation during infant or early childhood should be indicated for those patients with certain MYH14 pathogenic variants.
