Byung-Ok Choi 교수의 연구실은 신경병증과 근병증의 유전적 기반을 규명하는 데 초점을 맞추고 있으며, 특히 색소매병(CMT)과 청소년기 발병 근이영양증을 비롯한 퇴행성 신경근질환의 유전자 원인을 밝혀내는 데 전문성을 가진다. 전장 엑소좀 시퀀싱을 활용한 희귀 유전질환의 유전자 진단 및 신규 변이 탐색이 핵심 연구 전략이며, 임상과 기초를 융합한 개인 맞춤형 진료 기반의 유전체 연구를 추진하고 있다. 특히 PMP22, PMP2, DGAT2, ADSSL1 등 다양한 유전자에서의 새로운 병원성 변이를 규명하며 유전적 이질성과 임상 양상 간의 관계를 탐색하고 있다.

We examined CMT1A duplication of 17p11.2-p12, mutations of PMP22, MPZ (P0), GJB1 (Cx32), EGR2 and NEFL genes in 57 Korean families with patients diagnosed as having Charcot-Marie-Tooth (CMT) disease. The CMT1A duplication was present in 53.6% of 28 CMT type 1 patients. In the 42 CMT families without CMT1A duplication, 10 pathogenic mutations were found in 9 families. Both peripheral neuropathy and distal myopathy are well-established inherited neuromuscular disorders characterized by progressive weakness and atrophy of the distal limb muscles. A complex phenotype of peripheral neuropathy, myopathy, hoarseness, and hearing loss was diagnosed in a large autosomal dominant Korean family. Charcot-Marie-Tooth disease (CMT) is a heterogeneous group of peripheral neuropathies with diverse genetic causes. We suggest that mutations in ADSSL1 are the novel genetic cause of the autosomal recessive adolescent onset distal myopathy. This study broadens the genetic and clinical spectrum of distal myopathy and will be useful for exact molecular diagnostics.

Mutations of the mitofusin 2 (MFN2) gene have been reported to be the most common cause of the axonal form of Charcot-Marie-Tooth disease (CMT). A prospective brain MRI study was performed on 18 early-onset CMT patients with MFN2 mutations, and a high frequency (39%) of brain abnormalities was found.
