Do Sik Min 교수의 연구실은 세포 신호전달에서 핵심적인 역할을 하는 인지질분해효소인 인산지질분해효소D(PLD)의 활성화 메커니즘과 그 기능을 중심으로 연구를 진행하고 있습니다. 특히 PLD의 티로신 인산화, c-Src 및 성장인자 수용체와의 상호작용, 그리고 암 발생 및 신경퇴행성질환과의 연관성에 중점을 두고 있습니다. 또한 PLD의 조절 메커니즘과 그 기능적 영향을 다양한 세포 모델을 통해 규명하고 있습니다.

We have investigated the mechanisms involved in H2O2-mediated phospholipase D (PLD) activation in Swiss 3T3 fibroblasts. In the presence of vanadate, H2O2 induced tyrosine phosphorylation of PLD as well as the platelet-derived growth (PDGF) factor receptor, protein kinase Calpha (PKCalpha), and a 62-kDa protein in rat brain PLD1 (rPLD1) immune complexes. Phospholipase D (PLD) has been implicated in the signal transduction pathways initiated by several mitogenic protein tyrosine kinases. We have recently cloned a cDNA encoding a phospholipase D (PLD) from rat brain and named it rPLD1. It shows 90% amino acid identity with the human PLD isoform hPLD1b. Phospholipase D (PLD) has been suggested to play an important role in a variety of cellular functions. PLD activity has been shown to be significantly elevated in many tumours and transformed cells, suggesting the possibility that PLD might be involved in tumorigenesis. In this study, we have established stable cell lines overexpressing PLD1 and PLD2 from fibroblast cells.
