Proteins are fundamentally the most important macromolecules for biochemical, mechanical, and structural functions in living organisms. Therefore, they provide us with diverse structural building blocks for constructing various types of biomaterials, including an important class of such materials, hydrogels. Since natural peptides and proteins are biocompatible and biodegradable, they have features advantageous for their use as the building blocks of hydrogels for biomedical applications. They d
A growing appreciation of the relationship between the immune system and the tumorigenesis has led to the development of strategies aimed at "re-editing" the immune system to kill tumors. Here, a novel tactic is reported for overcoming the activation-energy threshold of the immunosuppressive tumor microenvironment and mediating the delivery and presentation of tumor neoantigens to the host's immune system. This nature-derived nanocage not only efficiently presents ligands that enhance cancer cel
Considering the problems of small interfering RNA (siRNA) delivery using traditional viral and nonviral vehicles, a new siRNA delivery system to enhance efficiency and safety needs to be developed. Here human ferritin‐based proteinticles are genetically engineered to simultaneously display various functional peptides on the surface of proteinticles: cationic peptide to capture siRNA, tumor cell targeting and penetrating peptides, and enzymatically cleaved peptide to release siRNA inside tumor ce
Desertification is a global phenomenon caused by various processes, including climate change, vegetation processes, and human activities. The need to combat desertification is increasing in many countries. A reasonable assessment of the vulnerability or sensitivity of land cover to desertification at national scales is crucial to formulate appropriate strategies or policies for combating it. The main purpose of this work was to quantitatively assess the sensitivity of land cover to desertificati
Xylose, the second most abundant sugar in lignocellulosic biomass hydrolysates, can be fermented by Saccharomyces cerevisiae expressing one of two heterologous xylose pathways: a xylose oxidoreductase pathway and a xylose isomerase pathway. Depending on the type of the pathway, its optimization strategies and the fermentation efficiencies vary significantly. In the present study, we constructed two isogenic strains expressing either the oxidoreductase pathway (XYL123) or the isomerase pathway (X
Although the importance of fluid flow for proper vascular development and function in vivo is well recognized, microvascular formation in response to flow has not been well evaluated in a three-dimensional (3D) environment in vitro. In this study, we developed a novel 3D in vitro perfusion system that allows direct investigation of the effects of shear stress on the development of microvasculature in vitro. This system utilizes a 3D collagen gel for suspension of vascular cells and mesenchymal s
Ferritin-based nanoprobe (FBNP) hydrogel provides solutions to some of the traditional problems of bioassays. The schematic shows 3D and highly specific multiplex assays of markers for AIDS and Sjögren's syndrome using quantum-dot based reporters with different fluorescence emissions for the two syndromes. The advantages of using a hydrogel include that the stability of FBNPs is significantly enhanced within the hydrogel, FBNPs are distributed evenly throughout the hydrogel matrix, and the amoun
"Proteinticle" is a nano-scale protein particle that is self-assembled inside cells with constant 3D structure and surface topology. The binding of IgG to the B domain of Staphylococcal protein A (SPA(B)) molecules that are genetically inserted on the surface of proteinticle enables the variable domains of bound IgG to be well oriented to effectively capture antigens, accordingly forming a highly sensitive 3D IgG probe. The five different proteinticles that originate from humans, bacteria, and v
Tumor-specific apoptosis-inducing ligands have attracted considerable attention in cancer therapy. But, the evasion of apoptosis by tumors can cause acquired resistance to the therapy. TNF-related apoptosis-inducing ligand (TRAIL) has been investigated as an ideal antitumor agent owing to its inherent tumor cell-specific apoptotic activity. However, there are several barriers to its wider application, including the inability for stable formation of the trimeric structure, poor stability and phar
Abstract To date, only a few nanosystems have been investigated as T 1 MRI‐CT dual contrast agents. The T 1 MRI‐CT dual functionality of a material depends on its longitudinal water‐proton relaxivity ( r 1 ) and X‐ray absorption strength. We explored Gd(IO 3 ) 3 · 2H 2 O nanomaterial because Gd is the most powerful element for T 1 MRI contrast agents, and both Gd and I absorb X‐ray radiation; Gd absorbs X‐ray radiation ca. 2.5 times more strongly than I. D ‐Glucuronic acid coated Gd(IO 3 ) 3 · 2
The pandemic caused by the severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) has upended healthcare systems and economies around the world. Rapid understanding of the structural biology and pathogenesis of SARS-CoV-2 has allowed the development of emergency use or FDA-approved vaccines and various candidate vaccines. Among the recently developed SARS-CoV-2 candidate vaccines, natural protein-based nanoparticles well suited for multivalent antigen presentation and enhanced immune stimu
