Mechanical stresses on the myocyte nucleus have been associated with several diseases and potentially transduce mechanical stimuli into cellular responses. Although a number of physical links between the nuclear envelope and cytoplasmic filaments have been identified, previous studies have focused on the mechanical properties of individual components of the nucleus, such as the nuclear envelope and lamin network. The mechanical interaction between the cytoskeleton and chromatin on nuclear deform
We demonstrate a stochastic based method for performing active rheology using optical tweezers. By monitoring the displacement of an embedded particle in response to stochastic optical forces, a rapid estimate of the frequency dependent shear moduli of a sample is achieved in the range of 10(-1)-10(3) Hz. We utilize the method to probe linear viscoelastic properties of hydrogels at varied cross-linker concentrations. Combined with fluorescence imaging, our method demonstrates non-linear changes
Collagen is the most abundant extracellular matrix protein. The concentrations, structural arrangement, and directionality of collagen depend on the type of tissue. Thick fibril bundles of collagen are observed in most collagenous tissues, including connective tissues, bones, and tendons, indicating that they play a critical role in many cell functions. In this study, we developed a new method to regulate collagen bundling without altering the protein concentration, temperature, or pH by using s
Two types of measurement are presented that relate molecular events to macroscopic behavior of F-actin networks. First, shear modulus is measured by oscillating an embedded microbead. Second, a microbead is translated at constant rate and transitions in the resisting force are observed. The loading rate dependence of the force at the transitions is similar to that of the molecular unbinding force, suggesting that they share a common origin. Reversibility tests of shear modulus provide further ev
There is a growing demand in the development of small-scale devices in microelectronics and microelectromechanical systems (MEMS). Packaging and reliability of such devices are of great concern as they introduce a number of unique packaging issues that are distinct and different from typical electronic packaging applications. In addition, the packaging or encapsulation materials are often exposed to harsh environments, for which their performance is drastically degraded. Importantly, such device
Standing surface acoustic wave (SSAW) combined with a microfluidic channel has been emerging as a device to manipulate biological and chemical particles for separation, mixing, and enrichment. Particles are trapped at pressure minima in the interference pattern determined by the acoustic properties of the fluid and the channel wall. To predict particle motion, it is essential to analyze the pressure distribution formed in the fluid. Here, we investigated the effect of sound speeds of fluid and c
Matrix stiffness, a critical physical property of the cellular environment, is implicated in epidermal homeostasis. In particular, matrix stiffening during the pathological progression of skin diseases appears to contribute to cellular responses of keratinocytes. However, it has not yet elucidated the molecular mechanism underlying matrix-stiffness-mediated signaling in coordination with chemical stimuli during inflammation and its effect on proinflammatory cytokine production. In this study, we
The lateral resolution of continuous wave (CW) stimulated emission depletion (STED) microscopy is enhanced about 12% by applying annular-shaped amplitude modulation to the radially polarized excitation beam. A focused annularly filtered radially polarized excitation beam provides a more condensed point spread function (PSF), which contributes to enhance effective STED resolution of CW STED microscopy. Theoretical analysis shows that the FWHM of the effective PSF on the detection plane is smaller
