오오타 쿠니히로 교수의 연구실은 세포 분열과 유전자 재조합을 둘러싼 세포골격 및 염색체 구조의 분자 기전을 중심으로 연구를 진행하고 있습니다. 특히 미세소관의 형성과 조절, 세포 분열에서의 중심체 기능, 그리고 면역글로불린 유전자에서 관찰되는 유전자 전환 메커니즘 등 유전자 다양성과 유전자 공통진화를 규명하는 데 초점을 맞추고 있습니다. 다양한 모델 생물(해면충, 효모, Xenopus)을 활용한 생화학적·세포생물학적 분석을 통해 세포 내 구조와 유전자 기능의 상호작용을 탐구하고 있습니다.

We investigated the biochemical characteristics of the 51-kDa protein that is a major mitotic apparatus-associated basic protein of sea urchin eggs (Toriyama, M., Ohta, K., Endo, S., and Sakai, H. In the yeast Saccharomyces cerevisiae, meiotic recombination is initiated by DNA double-stranded breaks (DSBs) occurring in micrococcal nuclease (MNase)-hypersensitive regions of the chromatin. Gene conversion is a type of homologous recombination that leads to transfer of genetic information among homologous DNA sequences. It can be categorized into two classes: homogenizing and diversifying gene conversions. We have studied the regulation of microtubule nucleating activity of the centrosome using cell-free extracts from Xenopus eggs. We found that the number of microtubules per centrosome increases dramatically with time during incubation of isolated centrosomes in interphasic egg extracts prepared 20-30 minutes after electric activation of cytostatic factor (CSF)-arrested eggs. The increase in microtubule nucleation was still conspicuous even when KCl-treated centrosomes (centrosomes stripped of th
