Kyoung-Ho Pyo 교수의 연구실은 암의 대사 재프로그래밍과 O-갈락토사미닐레이션(OGT)을 중심으로 한 단백질 수식 전환 메커니즘을 규명하며, 암의 진행과 전이를 조절하는 분자 기전을 연구하고 있습니다. 특히, 기생충 기반 면역 조절제인 톡소플라isma gondii 유전자 단백질(예: TgPLP, TLA)이 암 미세환경을 변화시켜 종양 성장과 혈관신생을 억제하고, 면역세포 기능을 향상시키는 작용을 통해 암 치료의 새로운 전략을 모색하고 있습니다. 또한, 면역검사점 억제제에 내성을 보이는 암에서 SKI-G-801과 같은 신규 면역 조절제가 저항성을 극복할 수 있는 가능성을 탐색하고 있습니다.

Toxoplasma gondii is an intracellular protozoan parasite that causes a Th1 cellular immunity. Our previous study showed that T. gondii lysate antigen (TLA) treatment in S180 tumor-bearing mice resulted in tumor reduction by suppressing CD31 expression, a marker of angiogenesis. SKI-G-801 significantly suppressed tumor metastasis and growth by enhancing anti-tumor immune responses. Our results suggest that SKI-G-801 has the potential to overcome anti-PD-1 therapy resistance and allow more patients to benefit from anti-PD-1 therapy.
These findings highlight the multifaceted roles of CD81 and CD82 in T cell activation, cytokine production, memory subset accumulation, and target cell cytolysis. Therefore, these findings suggest the potential of CD81 and CD82 as promising candidates for co-stimulatory molecules in immune therapeutic strategies for cancer treatment within the intricate TME.

The activation of macrophages by trypsin-digested silk peptides was investigated by considering CD1 lb and CD40 expression in the RAW264.7 cell, a murine macrophage. Silk protein hydrolysates were digested with trypsin, following by centrifugal purification using the Centriprep 30k concentrator. Trypsin-digested total silk peptides and its centrifugal fractions were tested for macrophage activation in vitro. The functional peptide of fractionated silk peptides was examined by LC/MS/MS analysis.
