Shigeo Okabe 교수의 연구실은 신경세포의 구조적 및 기능적 다이나믹스를 분자생물학적·영상 기술을 융합하여 연구합니다. 특히 시냅스 형성과 기능, 미세소관과 액틴 필라멘트의 동적 재편성, 신경세포 성장공의 모양 변화 메커니즘을 중심으로 신경 회로의 기초를 밝혀내고자 합니다. 고해상도 시간-공간 분辨 영상 기술을 활용해 단백질의 위치와 이동을 실시간으로 관찰함으로써, 뇌 기능의 세포기반 기전을 해명하는 데 초점을 맞추고 있습니다.

Hippocampal pyramidal neurons in culture showed a developmental shift in synapse distribution from dendritic shafts to spines. To study microtubule (MT) dynamics in nerve cells, we microinjected biotin-labeled tubulin into the cell body of chemically fused and differentiated PC12 cells and performed the immunofluorescence or immunogold procedure using an anti-biotin antibody followed by secondary antibodies coupled to fluorescent dye or colloidal gold. PSD-Zip45 (Homer 1c) and PSD-95 are postsynaptic density (PSD) proteins containing distinct protein-interacting motifs. Green fluorescent protein (GFP)-tagged PSD-Zip45 and PSD-95 molecules were targeted to the PSD in hippocampal neurons. The effects of changing NMDA receptor subunit composition on synaptic plasticity in the hippocampus were analyzed by creating transgenic mice overexpressing NR2D, a predominantly embryonic NMDA receptor subunit.
