우치다 타츠야 교수의 연구실은 페리린기반 촉매를 중심으로 한 고선택성 산화 반응 및 C–H 기능화 기반의 유기합성 반응을 연구하고 있습니다. 특히 루테늄-살렌 복합체를 이용한 빛 활성화 반응을 통해 사이클로프로판화, 아지리딘화, 아미노기 기능화 등 고대칭성과 고 diastereo-또는 enantioselectivity를 확보한 반응을 개발하고 있습니다. 이는 약물 합성 및 자연물 합성에서 중요한 기능기 도입 전략으로 응용 가능합니다.

Nitrogen functional groups are found in many biologically active compounds and their stereochemistry has a profound effect on biological activity. The asymmetric nitrene transfer reaction is a useful and strong tool for the construction of nitrogen functional groups such as <i>N-</i>sulfonyl amide and carbamic ester in a highly enantioselective manner. The reaction of styrene and t-butyl α-diazoacetate in the presence of (R,R)-(ON+)(salen)ruthenium(II) complex 1 under the irradiation of incandescent light in THF gave the corresponding (1S,2R)-cyclopropanecarboxylate with high stereoselectivity of 99% ee (cis : trans = 96 : 4), while the same reaction in hexane gave the enantiomeric (1R,2S)-cyclopropanecarboxylate preferentially (83% ee, cis : trans = 68 : 32).

If such C–H bonds can be converted into the desired functional groups in a site-, chemo-, diastereo-, and enantio-selective manner, the functionalization of C–H bonds would be an efficient tool for step-, atom- and redox-economic organic synthesis.
