Tomoo Ogi 교수의 연구실은 DNA 손상 복구와 변이 발생 메커니즘을 중심으로, 특히 전이손상 합성(Translesion Synthesis, TLS)을 담당하는 DNA 폴리메라아제의 기능과 regulation을 연구하고 있습니다. 특히 벤조[a]피렌 등 환경 유발 유전자 손상에 대한 정확한 복구 메커니즘과, 이와 관련된 유전자 변이가 유전성 질환 및 암 발생에 미치는 영향을 규명하고 있습니다. 또한, DNA 복구 경로와 대사 경로의 상호작용, 예를 들어 알코올 대사 유전자와 Fanconi 빈혈 경로의 연관성 등, 복합적인 생물학적 네트워크를 탐구하고 있습니다.

Several low-fidelity DNA polymerases have recently been discovered that are able to bypass DNA lesions during DNA synthesis in vitro. The above results suggest that a mutagenic mechanism, a so-called untargeted type, also operates in mammalian cells. Taken together with recent findings that human cells have multiple DNA polymerases for translesion synthesis which are homologous to the S. cerevisiae Rev3 and Rad30 proteins, our results imply that multiple mutagenic pathways are conserved from bacteria to higher eukaryotes.
Xeroderma pigmentosum (XP) is an autosomal recessive genetic disorder. Afflicted patients show extreme sun-sensitivity and skin cancer predisposition. The AhR activation increases expression of the mouse Polk gene and probably the human POLK gene, the product of which bypasses benzo[a]pyrene-N2-dG lesions in a mostly accurate manner.
