The binding mode of CA074 [N-(L-3-trans-propylcarbamoyl-oxirane-2-carbonyl)-L-isoleucyl-L-pr oline], a specific irreversible inhibitor, to bovine spleen cathepsin B was elucidated by X-ray crystal structure analysis of the complex at 2.2 A resolution (conventional R=0.185). Inconsistently with our model used for the development of CA074, the L-isoleucyl-L-proline and propylcarbamoyl moieties are located at the S' and S subsites, respectively. This unexpected binding is primarily due to (i) simil

The effects of changes in the anterior-posterior center of pressure (AP-COP) position on the lower limb joint moments during double-leg squatting remain unclear. The purpose of this study was to determine the effects of AP-COP positional changes on the hip, knee, and ankle extensor moments during double-leg squatting. Sixteen male participants (22.1 ± 1.5 years) performed double-leg squatting under two conditions (anterior and posterior COP conditions) with visual feedback on their COP positions
Subsequent jumps cause greater knee abduction during the early landing phase only in female subjects. This finding may relate to the sex discrepancy in non-contact anterior cruciate ligament injuries. Additionally, the presence of a subsequent jump significantly increases the peak knee abduction angle and the peak knee abduction moment during landings. Therefore, compared with a landing task without a subsequent jump (drop landing), a landing task with a subsequent jump (drop vertical jump) may
The AP-COP position can predict the contribution of the knee extensor moment and improve the prediction when combined with ankle dorsiflexion and trunk flexion. The present findings suggest that intervention focusing on the AP-COP position in combination with ankle dorsiflexion or trunk flexion would be useful to coordinate the contribution of the knee extensor moment during double-leg squatting.
The ACL-RSI score was significantly different among the study groups, and the ACL-RSI score at 12 months postoperatively and younger age were associated with RTS at or above preinjury level.
Associations among hip motions, knee abduction and internal rotation motion during a drop vertical jump (DVJ), which increases the risk of anterior cruciate ligament injury, remain unclear. The purpose of this study was to examine associations among knee abduction, internal rotation and hip joint motions during a DVJ. Fifty-seven young female participants performed a DVJ from a 30-cm height. Hip and knee kinematics and kinetics were analysed using a three-dimensional motion analysis system and f
We assessed the accuracy of deformable image registration (DIR) accuracy between CT and MR images using an open-source software (Elastix, from Utrecht Medical Center) and a commercial software (Velocity AI Ver. 3.2.0 from Varian Medical Systems, Palo Alto, CA, USA) software. Five male patients' pelvic regions were studied using publicly available CT, T1-weighted (T1w) MR, and T2-weighted (T2w) MR images. In the cost function of the Elastix, we used six DIR parameter settings with different regul
Knee kinematics during gait is an important assessment tool in health-promotion and clinical fields. This study aimed to determine the validity and reliability of a wearable goniometer sensor for measuring knee flexion angles throughout the gait cycle. Twenty-two and seventeen participants were enrolled in the validation and reliability study, respectively. The knee flexion angle during gait was assessed using a wearable goniometer sensor and a standard optical motion analysis system. The coeffi
This study investigated the relationship between quadriceps strength and knee kinematics during a drop vertical jump (DVJ) at 6, 9 and 12 months after anterior cruciate ligament reconstruction (ACLR) in 9 male and 22 female athletes (16.6 ± 2.1 years old). Isokinetic quadriceps strength was measured by a dynamometer (Biodex System 3). Knee flexion excursion was assessed using two-dimensional analysis. Knee flexion excursion at 6 months was significantly smaller in the involved limb than in the u
Quadriceps strengthening exercises, together with interventions to improve neuromuscular control, may reduce asymmetrical biomechanics during double-leg squatting.
