노자키 도모요시 교수의 연구실은 주로 기생성 원생동물, 특히 아메바성 기생충 엔테로아미يب아 히스톨리티카의 세포 내 구조와 기능, 특히 미토콘드리아 유사기관(MROs), 포식체 생합성, 철황소(Fe-S) 클러스터 형성 및 단백질 분포 메커니즘을 중심으로 연구를 진행하고 있습니다. 특히 미토콘드리아 기원의 기능이 손실된 환경에서 어떻게 세포 기능이 유지되는지, 그리고 병원성 기생충의 대사 경로와 세포 신호 전달 체계의 기초를 밝히는 데 초점을 맞추고 있습니다. 이는 기생충의 병원성 기전 규명과 새로운 약물 타겟 탐색에 기여할 수 있습니다.

In mammals, Rab5 and Rab7 play a specific and coordinated role in a sequential process during phagosome maturation. Here, we report that Rab5 and Rab7 in the enteric protozoan parasite Entamoeba histolytica, EhRab5 and EhRab7A, are involved in steps that are distinct from those known for mammals. The enteric protist parasites Entamoeba histolytica and Entamoeba dispar possess a cysteine biosynthetic pathway, unlike their mammalian host, and are capable of de novo production of L-cysteine. Proteomic analysis of phagosomes isolated from Entamoeba histolytica by liquid chromatography and mass spectrometry identified 85 proteins involved in surface recognition, actin cytoskeleton rearrangement, vesicular trafficking, and degradation. Phagosome localization of representative proteins was verified by immunofluorescence assay. This study should provide a basis for molecular identification and characterization of phagosome biogenesis.
We have characterized the iron-sulfur (Fe-S) cluster formation in an anaerobic amitochondrial protozoan parasite, Entamoeba histolytica, in which Fe-S proteins play an important role in energy metabolism and electron transfer.
