토요아키 무로하라 교수의 연구실은 혈관 형성 및 혈관 내피 기능 조절 메커니즘을 중심으로 연구를 진행하고 있습니다. 특히 염증성 혈관성장 인자(VEGF)와 일산화질소(NO)의 상호작용이 혈관형성과 혈관투과성에 미치는 영향을 규명하며, 내피선구자세포(EPC)의 기능과 이식을 통한 혈관신생 조절 메커니즘을 탐구하고 있습니다. 이와 더불어 L-아르기닌을 통한 NO 생성 증강이 허혈성 조직에서의 혈관신생을 어떻게 촉진하는지에 대한 기전 연구도 진행 중입니다.

We tested the hypothesis that endothelial nitric oxide synthase (eNOS) modulates angiogenesis in two animal models in which therapeutic angiogenesis has been characterized as a compensatory response to tissue ischemia. We first administered L-arginine, previously shown to augment endogenous production of NO, to normal rabbits with operatively induced hindlimb ischemia. These results implicate NO and prostacyclin produced by the interaction of VEGF/VPF with its Flk-1/KDR/VEGF-R2 receptor as mediators of VEGF/VPF-induced vascular permeability. Moreover, this property appears unique to VEGF/VPF among angiogenic cytokines.
Endothelium-derived nitric oxide (NO) and its precursor L-arginine have been implied to promote angiogenesis, but little is known about the precise mechanism.
