Won-Il Jeong 교수의 연구실은 간 섬유화의 분자 기전과 그 치료적 타겟을 규명하는 데 초점을 맞추고 있습니다. 주로 간성성세포(hepatic stellate cells)의 활성화, TLR3-IL-17A 신호 경로, IFN-γ 및 NK 세포의 항섬유화 작용, 그리고 미토콘드리아 유래 RNA(mtdsRNA)가 유도하는 염증 반응을 중심으로 연구를 진행하고 있습니다. 특히 알코올성 간질환과 고콜레스테롤 식이에 의한 간 손상 모델을 활용해 간 섬유화의 병태생리학적 기전을 체계적으로 규명하고 있습니다.

Liver fibrosis, a common scarring response to chronic liver injury, is a precursor to cirrhosis and liver cancer. Here, we identified signal transducer and activator of transcription 1 (STAT1) as an important negative regulator in liver fibrosis. In liver injury, the exosome-mediated activation of TLR3 in HSCs exacerbates liver fibrosis by enhancing IL-17A production by γδ T cells, which might be associated with HSC stimulation by unknown self-TLR3 ligands from damaged hepatocytes. Therefore, TLR3 might be a novel therapeutic target for liver fibrosis. (Hepatology 2016;64:616-631).
The antifibrogenic effects of NK cell/IFN-γ are suppressed during advanced liver injury, which is likely due to increased production of TGF-β and expression of SOCS1 in intermediately activated HSCs.
EtOH-mediated generation of mtdsRNA contributes to TLR3 activation in Kupffer cells through exosomal delivery. Consequently, increased IL-1β expression in Kupffer cells triggers IL-17A production in γδ T cells at the early stage that may accelerate IL-17A expression in CD4<sup>+</sup> T cells in the later stage of ALD. Therefore, mtdsRNA and TLR3 may function as therapeutic targets in ALD.

Three dead dogs were brought to the College of Veterinary Medicine, Kyungpook National University for study. Clinically, all the dogs showed emaciation, anorexia, depression, hemorrhagic vomiting and diarrhea for 7-10 days before death. The numbers of macrophages and myofibroblasts increased in the development of fibrosis but decreased when cirrhosis was induced. However, mast cells increased in both. Therefore, there was a numerical relationship among the numbers of macrophages, myofibroblasts and mast cells in liver fibrosis but none between the former two cells and the mast cells in liver cirrhosis.
