microRNAs (miRNAs) play integral roles in diverse processes including tumorigenesis. miRNA gene loci are often found in close conjunction, and such clustered miRNA genes are transcribed from a common promoter to generate polycistronic primary transcript. Biogenesis of canonical microRNAs (miRNAs) involves multiple steps: nuclear processing of primary miRNA (pri-miRNA) by DROSHA, nuclear export of precursor miRNA (pre-miRNA) by Export in 5 (XPO5), and cytoplasmic processing of pre-miRNA by DICER. The structures of pyripyropenes A, B, C and D, novel acyl-CoA:cholesterol acyltransferase (ACAT) inhibitors, were determined mainly by spectroscopic studies including various NMR measurements. Pyripyropenes have a common structure which consists of pyridine, alpha-pyrone and sesquiterpene moieties. One of the three O-acetyl residues in the sesquiterpene moiety of pyripyropene A is replaced with an O-propionyl residue in pyripyropenes B, C and D.
[reaction: see text] A novel trifluoroacetophenone-based binding motif has been developed that recognizes anions such as carboxylates through reversible formation of anion-ionophore adducts that are stabilized by intramolecular H-bonding. The intramolecular H-bonding resulted in more than 10-fold enhancement in the binding affinity and an enthalpy gain (DeltaH degrees ) of 3.0 kcal/mol for the binding of an acetate ion when compared to the case without the intramolecular H-bonding.
Three new hybrid ligands with trans-1,2-diaminocyclohexane backbone have been synthesized from (1R,2R)-2-aminocyclohexylcarbamic acid tert-butyl ester (4), which is prepared through an indirect monoprotection of the diamine. Focal LC defects are more frequently visible in eyes with glaucomatous DH than in eyes without DH. DH and LC defects, moreover, are often spatially correlated. These findings suggest that DH is associated with focal abnormalities of the LC.
