Yuichi Riku 교수의 연구실은 신경퇴행성질환, 특히 TDP-43 및 타우 단백질 관련 퇴행성 병변의 분자 및 병리학적 기전을 중심으로 연구를 진행하고 있습니다. 아밀로이드성 뇌병변, 파킨슨병, 프리온병 등 다양한 퇴행성 뇌질환에서 TDP-43와 타우의 공존 병변이 어떻게 발생하고 진행되는지를 밝혀내는 데 초점을 맞추고 있으며, 유전적 변이와 병리학적 소견 간의 연관성도 깊이 연구하고 있습니다. 특히 ALS, FTLD-TDP, tauopathy 등에서의 신경계 축삭과 운동 뉴런 손상의 병리학적 연속성을 규명하고 있습니다.

PMA has three neuropathological background patterns. A combination of UMN and LMN degeneration with TDP-43 pathology, consistent with ALS, is the major pathological profile. The remaining patterns have LMN degeneration with TDP-43 pathology without UMN degeneration, or a combination of UMN and LMN degeneration with FUS-positive basophilic inclusion body disease.
Our results support a genetic clinical view that compound heterozygous mutations with potential residual protein function are associated with a relatively mild phenotype. Moreover, the severe LB pathology suggests that dysfunction of the PLA2G6 gene primarily contributes to LB formation.
The LMN systems of FTLD-TDP frequently exhibit neuropathological changes corresponding to ALS. Thus, a pathological continuity between FTLD-TDP and ALS is supported at the level of the LMN system.

Amyotrophic lateral sclerosis (ALS) is a motor neuron disease that is clinically and pathologically characterized by impairment of the upper and lower motor neurons.
