Chikashi Toyoshima教授の研究室は、膜タンパク質の構造生物学に特化しており、特にカルシウムポンプ(SERCA1a)の構造と機能の解明を主眼としています。X線結晶構造解析と低温電子顕微鏡を用いて、イオン輸送のメカニズムやリン酸化状態、調節因子(例如:プロスタグランジン)との相互作用を原子レベルで解明しています。特に、Ca²⁺結合状態と非結合状態の構造的変化、およびATP加水分解サイクルにおけるドメイン間の力学的連動を解明しています。

The structures of the Ca2+-ATPase (SERCA1a) have been determined for five different states by X-ray crystallography. Detailed comparison of the structures in the Ca2+ bound form and unbound (but thapsigargin bound) form reveals that very large rearrangements of the transmembrane helices take place accompanying Ca2+ dissociation and binding and that they are mechanically linked with equally large movements of the cytoplasmic domains. Long tubular vesicles have been grown from isolated Torpedo postsynaptic membranes, in which the receptors are arranged helically on the vesicle surface.
