Professor Do Hyun Ryu's research lab specializes in the development and application of chiral oxazaborolidine- and oxazaborolidinium-based catalysts for asymmetric synthesis. The lab focuses on designing highly enantioselective transition-metal-free catalytic systems, particularly for Diels-Alder reactions, cyanosilylation, and Morita-Baylis-Hillman-type transformations. Key advances include the rational design of superactive chiral Lewis acids through triflimide or triflic acid activation, enabling predictable facial selectivity and high enantioselectivity across diverse substrates. The strong acid triflimide ((CF3SO2)2NH) protonates chiral oxazaborolidines to form superactive, stable, chiral Lewis acids which are highly effective catalysts for a wide variety of enantioselective Diels-Alder reactions, documented herein by more than 20 examples.
The cationic chiral Lewis acids 1 and 2, generated by triflic acid protonation of the corresponding neutral oxazaborolidines, serve as excellent catalysts for Diels-Alder addition of cyclopentadiene to a wide variety of dienophiles. The chiral oxazaborolidinium salt 1 (X = TfO) is an excellent catalyst for the cyanosilylation of methyl ketones promoted by trimethylsilyl cyanide and diphenylmethyl phosphine oxide as co-reactants (to generate Ph(2)MePOTMS(N=C:) as a reactive intermediate). A catalytic route toward chiral Morita-Baylis-Hillman esters by asymmetric coupling between alpha,beta-acetylenic esters, aldehydes, and trimethylsilyl iodide has been developed (see scheme). The reaction proceeds with high to excellent enantioselectivities, and the products can be transformed into beta-branched derivatives in a single step and with excellent retention of configuration. A catalytic route to highly functionalized chiral 2-pyrazolines by an asymmetric 1,3-dipolar cycloaddition reaction of ethyl diazoacetate with alpha-substituted and alpha,beta-disubstituted acroleins has been developed; in the presence of chiral (S)-oxazaborolidinium ion as catalyst, the reaction proceeded with high to excellent enantioselectivities (up to 99% ee).
Chiral Lewis acid catalyzed asymmetric formation of cyclobutanones from α-silyloxyacroleins and α-alkyl or α-aryl diazoesters has been developed.
