Eiki Takimoto教授の研究室は、心筋の肥大・リモデリングおよび心機能障害の発症メカニズムに焦点を当て、特に一氧化窒素(NO)/cGMP/PKGシグナル経路とその調節因子(PDE5、RGS2、NOS3)の心臓における機能を解明しています。高血圧に起因する圧力負荷に対する心筋の適応反応や、βアドレナリン作動薬・サイクリックGMP代謝酵素との相互作用が心機能に与える影響を、遺伝子改変マウスや生体外系系の細胞実験を用いて解析しています。特に、cGMPシグナルの局所的制御が心不全の予防や治療にどう寄与するかを基盤から解明する研究が特徴です。

Cardiac pressure load stimulates hypertrophy, often leading to chamber dilation and dysfunction. ROS contribute to this process. Regulation of cardiac beta-adrenergic response by cGMP is specifically linked to a nitric oxide-synthesis/PDE-5-hydrolyzed pool signaling via protein kinase G. Natriuretic peptide stimulation achieves greater detectable increases in cGMP but not protein kinase G activity and does not modulate beta-adrenergic response.
