Goichi Beck教授の研究室は、神経変性疾患の病態メカニズム解明を主眼としており、特にインファンタイル・ネウロアキソンアル・ジストロフィー(INAD)やパーキンソン病の神経変性機構に焦点を当てた分子・細胞生物学的・神経病理学的研究を推進しています。iPLA2βの欠損が神経細胞内リン脂質の再編成障害と鉄代謝異常を引き起こすメカニズムの解明や、TDP-43の異常蓄積がドーパミン作動性ニューロンに与える影響についての研究が進んでいます。また、L-ドーパ誘因性ジスキネジーの神経回路メカニズムや、生体マーカーとしてのpα-Synの可能性についても、臨床的応用に向けた基盤研究を展開しています。

Infantile neuroaxonal dystrophy (INAD) is a fatal neurodegenerative disease characterized by the widespread presence of axonal swellings (spheroids) in the CNS and PNS and is caused by gene abnormality in PLA2G6 [calcium-independent phospholipase A(2)β (iPLA(2)β)], which is essential for remodeling of membrane phospholipids. Accumulation of abnormal transactivation response DNA-binding protein of 43 kDa (TDP-43) independently induces dopaminergic neuronal loss in the substantia nigra without Lewy pathology, and results in typical Parkinson's disease-like motor symptoms.
Results show that regulation of striatal cyclic nucleotides by phosphodiesterase 10A inhibition could be a useful therapeutic approach for l-dopa-induced dyskinesia, and therefore data support further studies of selective phosphodiesterase 10A inhibitors for PD therapy. © 2018 International Parkinson and Movement Disorder Society.
