Sasaki教授の研究室は、エピジェネティクス、特にDNAメチル化のメカニズムとその細胞周期・発生過程における役割を解明することを主眼としています。特に、プリマーリィステム細胞や卵母細胞における非CGメチル化の分布と制御機構、およびその関連酵素(DNMT3B、CDCA7、HELLSなど)の機能を、ゲノムスケールのメチルーム解析を用いて解明しています。また、ICF症候群をはじめとするエピジェネティクス異常疾患の病態解明にも貢献しています。

DNA methylation is an epigenetic modification that plays a crucial role in normal mammalian development, retrotransposon silencing, and cellular reprogramming. ICF syndrome is a rare autosomal recessive disorder characterized by immunodeficiency, centromeric instability, and facial anomalies.
