The present study investigated the expressional regulation of PpDAM5 and PpDAM6, two of the six peach (Prunus persica) dormancy-associated MADS-box genes, in relation to lateral bud endodormancy. PpDAM5 and PpDAM6 were originally identified as homologues of Arabidopsis SHORT VEGETATIVE PHASE/AGAMOUS-LIKE 24 identified in the EVERGROWING locus of peach. Furthermore, PpDAM5 and PpDAM6 have recently been suggested to be involved in terminal bud dormancy. In this study, seasonal expression analyses
This study describes a novel F-box protein gene in the S-locus of sour cherry (Prunus cerasus) and sweet cherry (P. avium). The gene showed an S-haplotype-specific sequence polymorphism and the expression was specific to pollen. Genomic DNA blot analysis of eight sweet cherry cultivars with the probe for the F-box protein gene under low stringency conditions yielded RFLP bands specific to the S-haplotypes of each cultivar. We discuss the possibility of the gene for the F-box protein being a cand
To understand the molecular basis of the endodormancy of buds of perennial plants, we searched for the genes that are expressed preferentially in endodormant lateral buds of the deciduous fruit tree japanese apricot ( Prunus mume Sieb. et Zucc.) using suppression subtractive hybridization with mirror orientation selection (SSH/MOS). We generated two SSH/MOS libraries containing gene pools that are expressed preferentially in endodormant buds in comparison with paradormant or ecodormant buds to s
This report demonstrates the presence of S -ribonucleases ( S -RNases), which are associated with gametophytic self-incompatibility (SI) in Prunus L., in styles of self-incompatible and self-compatible (SC) selections of tetraploid sour cherry ( Prunus cerasus L.). Based on self-pollen tube growth in the styles of 13 sour cherry selections, seven selections were SC, while six selections were SI. In the SI selections, the swelling of pollen tube tips, which is typical of SI pollen tube growth in
Most deciduous fruit trees cultivated in the temperate zone require a genotype-dependent amounts of chilling exposure for dormancy release and bud break. In Japanese apricot (Prunus mume), DORMANCY-ASSOCIATED MADS-box 6 (PmDAM6) may influence chilling-mediated dormancy release and bud break. In this study, we attempted to elucidate the biological functions of PmDAM6 related to dormancy regulation by analyzing PmDAM6-overexpressing transgenic apple (Malus spp.). We generated 35S:PmDAM6 lines and
This study characterizes the S6m-haplotype, a mutated S6-haplotype with an altered HindIII cut site, of sour cherry (Prunus cerasus). Inheritance and pollination studies of S-haplotypes from reciprocal crosses between 'Erdi Botermo' (EB; S4S6mSa) and 'Rheinische Schattenmorelle' (RS; S6SaSbSc) revealed that the S6m-haplotype conferred unilateral incompatibility with a non-functional pistil component and a functional pollen component. Expression analyses of S6-RNase and SFB6, a candidate gene for
Most rosaceous fruit tree species exhibit gametophytic self-incompatibility (GSI). During the last decade, much progress has been made in identifying and characterizing the S-locus genes that control the specificity of the self-incompatible (SI) reaction in the Rosaceae. The pistil and pollen determinants of S-specificity in the Rosaceae are a ribonuclease and an F-box protein, respectively. From allele-specific sequence polymorphism of S-specificity genes, molecular S-genotyping systems have be
Bud dormancy allows most deciduous fruit tree species to avoid injury in unsuitable environments, synchronize their annual growth, and adapt to a temperate zone climate. Because bud dormancy affects next season’s fruit production and vegetative growth, it is considered one of the most important physiological factors that control fruit production. Recent global climate changes require us to better understand the genetic factors regulating bud dormancy, especially those that induce dormancy releas
In this study, we investigated seasonal changes in protein profiles in dormant flower buds of Japanese apricot (Prunus mume Siebold Zucc.) cultivars 'Ellching', from subtropical Taiwan, and 'Nanko', from temperate Japan. One protein, isolated by two-dimensional polyacrylamide gel electrophoresis of flower bud extracts, was shown by peptide sequencing to be a dehydrin (the group of D-11 LEA (late embryogenesis-abundant) proteins). Patterns of dehydrin protein and transcript accumulation differed
In temperate deciduous fruit crops such as Prunus spp., bud endodormancy is an important physiological phase affecting the timing of blooming and subsequent fruit development. Japanese apricot (Prunus mume) bears unmixed flower buds, separate from vegetative buds, that bloom slightly more than a month before vegetative bud burst. Seasonal expression of Prunus mume DORMANCY ASSOCIATED MADS-box genes (PmDAMs) has previously been analyzed only in vegetative buds, with an association between these g
Annual growth cycles of perennial horticultural and forest woody plant species of temperate and boreal regions can be divided into two phases, namely the growth and dormancy phases. During the dormancy phase, vegetative and reproductive meristems undergo dormancy to withstand subzero winter temperatures. The dormant state of the plant organs can be defined classically as a non-growing latent state, where no visible growth occurs in meristem containing plants structures, such as seeds and buds. I
This report demonstrates the presence of S-RNases in style of self-incompatible Japanese plum (Prunus salicina Lindl.) and presents information about cDNA sequences encoding the S-RNases. Two stylar glycoprotein spots that were found in highly basic zone of 2D-PAGE gel with Mr of about 30kDa shared similarity in N-terminal amino acid sequence and immunological characteristics with other rosaceous S-RNases and appeared to be S-RNases of Japanese plum. Deduced amino acid sequences from the cDNA cl
