Professor Jin Sook Yoon's research lab focuses on the pathophysiology of Graves' orbitopathy (GO), with a central emphasis on understanding the roles of oxidative stress, inflammation, and fibrosis in orbital fibroblasts. The lab investigates natural compounds such as quercetin for their anti-inflammatory and anti-fibrotic effects, aiming to develop novel therapeutic strategies for GO. Additionally, the lab explores clinical ophthalmic conditions such as meibomian gland dysfunction in patients with ocular prostheses and rare complications of cosmetic procedures, including vascular occlusion from dermal fillers. Cigarette smoking is known to aggravate Graves' orbitopathy (GO) severity by enhancing adipogenesis. We investigated the effect of quercetin, an antioxidant, on adipocyte differentiation induced by cigarette smoke extract (CSE) in primary cultured orbital fibroblasts (OFs) from GO patients. Compared to paired normal eyelids, the eyelids with an ocular prosthesis exhibited significant signs of MGD. These results indicate that clinical assessment and management recommendations for MGD may benefit patients with prosthetic eyes, especially those with ocular discomfort.
miR-146a plays a role as a negative regulator in the production of TGF-β-induced fibrotic markers. Thus, miR-146a may be involved in the regulation of fibrosis in orbital fibroblasts from patients with GO.
Quercetin inhibited fibrotic markers and affected MMP-2 and MMP-9 activities in primary cell and orbital fat tissue cultures from GO at nontoxic concentrations. Our results support the potential use of quercetin for active inflammation and treatment or prevention of chronic fibrosis in GO.
Patients with Graves' ophthalmopathy had reduced health-related quality of life and were more likely to be depressed, especially those with a sight threatening condition or significant diplopia. It is important to identify these patients to provide the necessary psychological support.
Our results indicated that miR-27a and miR-27b inhibited adipogenesis in orbital fibroblasts from GO patients. Further studies are required to examine the potential of miR-27a and miR-27b as targets for therapeutic strategies.
MicroRNA 146a was upregulated by inflammatory stress in orbital fibroblasts. Our results indicated that miR-146a had a positive effect on the anti-inflammatory process. MicroRNA 146a may play a role in the regulation of inflammation in orbital fibroblasts, and may participate in the pathogenesis of GO.
