本研究室では、糖鎖修飾を制御する酵素反応の開発と、その応用を柱に、ネオグリコプロテインの効率的合成や、糖鎖代謝に関与する酵素の機能解明を進めています。特に、アーティフィシャル・プロトホルミエ菌由来のエンドオxy-N-アセチルグルコサミナダーゼ(Endo-A)を用いたトランスグリコシル化反応の最適化や、シュードスカローマス・ポムベの遺伝子機能解析を通じて、膜交通や細胞内小胞体系のメカニズムを解明しています。また、フィッションイーストを用いた遺伝子発現系の構築や、糖鎖修飾を介したタンパク質工学の応用にも注力しています。

We describe a novel method for the enzymatic synthesis of neoglycoproteins. Endo-beta-N-acetylglucosaminidase from Arthrobacter protophormiae (Endo-A) had high levels of transglycosylation activity. The enzyme activity of Endo-A was markedly increased by adding 4-L-aspartyl-glycosylamine (GlcNAc-Asn) to the reaction mixture. We have cloned the gene, vps34+, from the fission yeast Schizosaccharomyces pombe which encodes an 801 amino acid protein with phosphatidylinositol 3-kinase activity. We describe a simple and efficient procedure for transformation of Schizosaccharomyces pombe. Although various mutant strains of the fission yeast Schizosaccharomyces pombe exhibit non-sexual flocculation, little is known about the mechanistic basis for this phenomenon, nor have genes encoding the implicated flocculin been identified.
