伊東隆史教授の研究室は、T細胞リンパ腫や成人T細胞白血病(ATL)をはじめとするT細胞腫瘍の病態解明と新規治療戦略の開発を柱としています。特にCCクエン酸受容体4(CCR4)の機能とその腫瘍免疫逃避への役割に注目し、CCR4を標的にした抗体療法(モガムリズマブ)の臨床的有効性や、腫瘍微小環境における免疫調節機構を解明しています。また、腫瘍細胞の自己免疫逃避機構や、白血球の移動・浸潤メカニズムに関する基礎的・臨床的研究を統合的に行っています。

KW-0761 demonstrated clinically meaningful antitumor activity in patients with relapsed ATL, with an acceptable toxicity profile. Further investigation of KW-0761 for treatment of ATL and other T-cell neoplasms is warranted.

Adult T-cell leukemia/lymphoma (ATLL) patients are highly immunocompromised, but the underlying mechanism responsible for this state remains obscure. Recent studies demonstrated that FOXP3, which is a master control gene of naturally occurring regulatory T (Treg) cells, is expressed in the tumor cells from a subset of patients with ATLL. Since most ATLL cells express both CD4 and CD25, these tumors might originate from CD4(+)CD25(+)FOXP3(+) Treg cells, based on their phenotypic characteristics.
