Tatsuo Kurihara教授の研究室は、極限環境に適応した微生物、特に南極海域に生息する好コールド性バクテリア『シュワンエリア・リビングストンエンシスAc10』をモデル生物として、低温下でも効率的に代謝を維持する分子機構を解明しています。特にエイコサペンタエン酸(EPA)の生合成機構や、低温条件下でのタンパク質発現系の構築に注力しており、バイオテクノロジー的応用にも貢献しています。また、脱ハロゲン化酵素の機構解明や、細胞内膜小胞体輸送の分子機構についても、酵母や細菌を用いた分子生物学的手法を駆使して研究を進めています。

Shewanella livingstonensis Ac10, a psychrotrophic gram-negative bacterium isolated from Antarctic seawater, produces eicosapentaenoic acid (EPA) as a component of phospholipids at low temperatures. EPA constitutes about 5% of the total fatty acids of cells grown at 4 degrees C. The Sec23p/Sec24p complex functions as a component of the COPII coat in vesicle transport from the endoplasmic reticulum. Here we characterize Saccharomyces cerevisiae SEC24, which encodes a protein of 926 amino acids (YIL109C), and a close homologue, ISS1 (YNL049C), which is 55% identical to SEC24. A recombinant protein expression system working at low temperatures is expected to be useful for the production of thermolabile proteins. We constructed a low-temperature expression system using an Antarctic cold-adapted bacterium, Shewanella sp. strain Ac10, as the host. We evaluated the promoters for proteins abundantly produced at 4 degrees C in this bacterium to express foreign proteins.
