Soga教授の研究室は、キャピラリー電気泳動と質量分析法を組み合わせたハイブリッド分析技術を基盤とし、細胞内代謝物の包括的・定量的解析を追求しています。特に、電荷をもつ極性代謝物(アミノ酸、有機酸、リン酸エステルなど)の高感度・高スループット分析に特化しており、微生物(大腸菌、枯草菌など)の代謝応答や発生過程における代謝動態の解明を進めています。近年では、CE-TOFMSを用いた非標識代謝物の定性・定量法の開発や、代謝フラックスの再配分を解明するための包括的メタボローム解析手法の確立が目立ちます。

A new approach for the comprehensive and quantitative analysis of charged metabolites by capillary electrophoresis mass spectrometry (CE-MS) is proposed. Metabolites are first separated by CE based on charge and size and then selectively detected using MS by monitoring over a large range of m/z values. Analysis of cellular components at multiple levels of biological information can provide valuable functional insights. We performed multiple high-throughput measurements to study the response of Escherichia coli cells to genetic and environmental perturbations. Analysis of metabolic enzyme gene disruptants revealed unexpectedly small changes in messenger RNA and proteins for most disruptants.
