Umeharu Ohto教授の研究室は、リセプター・リガンド相互作用の構造生物学的解明を柱としており、特にToll様受容体(TLR)やNLRP3インフラマスなど、先天免疫応答を制御するタンパク質の構造と機能の解明を進めています。LPS認識のMD-2・TLR4複合体や、TLR8の活性化機構、NLRP3インフラマスの阻害機構など、病態関連の分子メカニズムを高精度な結晶構造解析によって解明しています。また、疾患関連SNPsの構造的影響や、酵素阻害剤の作用機構についても、創薬的応用を視野に入れた研究を展開しています。

Endotoxic lipopolysaccharide (LPS) with potent immunostimulatory activity is recognized by the receptor complex of MD-2 and Toll-like receptor 4.
