Ohno教授の研究室は、皮膚のバリア機能を形づくる膜脂質、特にシアー酸化型セラミド(acylceraamide)の生合成機構を解明する研究を主軸としています。特に、CYP4F22やPNPLA1といった関与する酵素の同定とその機能解明を通じて、皮膚障害や先天性 ichthyosis の分子メカニズムを解明しています。また、長鎖脂肪酸の延伸酵素(ELOVL)の基質特異性やスフィンゴリピッド代謝のメカニズムについても、生化学的・代謝的アプローチで解明を進めています。
