15Adipose tissue grows by two mechanisms: hyperplasia (cell number increase) and hypertrophy (cell size increase). Genetics and diet affect the relative contributions of these two mechanisms to the growth of adipose tissue in obesity. In this study, the size distributions of epididymal adipose cells from two mouse strains, obesity-resistant FVB/N and obesity-prone C57BL/6, were measured after 2, 4, and 12 weeks under regular and high-fat feeding conditions. The total cell number in the epididymal
Morphogenesis, spontaneous formation of organism structure, is essential for life. In the pancreas, endocrine α, β, and δ cells are clustered to form islets of Langerhans, the critical micro-organ for glucose homeostasis. The spatial organization of endocrine cells in islets looks different between species. Based on the three-dimensional positions of individual cells in islets, we computationally inferred the relative attractions between cell types, and found that the attractions between homotyp
Abstract We show that statistical criticality , i.e. the occurrence of power law frequency distributions, arises in samples that are maximally informative about the underlying generating process. In order to reach this conclusion, we first identify the frequency with which different outcomes occur in a sample, as the variable carrying useful information on the generative process. The entropy of the frequency, that we call relevance , provides an upper bound to the number of informative bits. Thi
Adipose cells are unique in the dynamism of their sizes, a requisite for their main function of storing and releasing lipid. Lipid metabolism is crucial for energy homeostasis. However, the regulation of lipid storage capacity in conditions of energy excess and scarcity is still not clear. It is not technically feasible to monitor every process affecting storage capacity such as recruitment, growth/shrinkage and death of individual adipose cells in real time for a sufficiently long period. Insulin-producing pancreatic islet b cells (b-cells) are destroyed, severely depleted or functionally impaired in diabetes. Therefore, replacing functional b-cell mass would advance clinical diabetes management. We have previously demonstrated the importance of Cdk4 in regulating b-cell mass. Cdk4-deficient mice display b-cell hypoplasia and develop diabetes, whereas b-cell hyperplasia is observed in mice expressing an active Cdk4R24C kinase. While b-cell replication appears to be the primary me
The thermodynamic uncertainty relation sets the minimal bound of the cost-precision tradeoff relation for dissipative processes. Examining the dynamics of an internally coupled system that is driven by a constant thermodynamic force, we, however, find that the tradeoff relation of a subsystem is not constrained by the minimal bound of conventional uncertainty relation. We made our point explicit by using an exactly solvable model of interacting oscillators. As the number ($N$) of interacting osc
Insulin is secreted in a pulsatile manner from multiple micro-organs called the islets of Langerhans. The amplitude and phase (shape) of insulin secretion are modulated by numerous factors including glucose. The role of phase modulation in glucose homeostasis is not well understood compared to the obvious contribution of amplitude modulation. In the present study, we measured Ca2+ oscillations in islets as a proxy for insulin pulses, and we observed their frequency and shape changes under consta
Deep learning methods have had outstanding performances in various fields. A fundamental query is why they are so effective. Information theory provides a potential answer by interpreting the learning process as the information transmission and compression of data. The information flows can be visualized on the information plane of the mutual information among the input, hidden, and output layers. In this study, we examine how the information flows are shaped by the network parameters, such as d
The islets of Langerhans, responsible for controlling blood glucose levels, are dispersed within the pancreas. A universal power law governing the fractal spatial distribution of islets in two-dimensional pancreatic sections has been reported. However, the fractal geometry in the actual three-dimensional pancreas volume, and the developmental process that gives rise to such a self-similar structure, has not been investigated. Here, we examined the three-dimensional spatial distribution of islets
Pancreatic islets can adapt to oscillatory glucose to produce synchronous insulin pulses. Can islets adapt to other oscillatory stimuli, specifically insulin? To answer this question, we stimulated islets with pulses of exogenous insulin and measured their Ca2+ oscillations. We observed that sufficiently high insulin (> 500 nM) with an optimal pulse period (~ 4 min) could make islets to produce synchronous Ca2+ oscillations. Glucose and insulin, which are key stimulatory factors of islets, modul
Maximum likelihood estimation (MLE) is fundamental to system inference for stochastic systems. In some generality, MLE will converge to the correct model in the infinite data limit. In the context of physical approaches to system inference, such as Boltzmann machines, MLE requires the arduous computation of partition functions summing over all configurations, both observed and unobserved. We present a conceptually transparent data-driven inference computation based on a reweighting of observed c
