Niiro教授の研究室は、好中球やモノサイトをはじめとする免疫細胞におけるプロスタグランジン産生の制御機構に注目し、特にIL-10やIL-4といった抗炎症サイトカインが炎症性メディエーターの産生をどのように抑制するかを分子メカニズムレベルで解明しています。特に、COX-2の遺伝子発現抑制を介したプロスタグランジンE2産生の制御が、炎症反応の調節において中心的な役割を果たすことが明らかになっています。この研究は、慢性炎症や自己免疫疾患の治療戦略の開発にも貢献しています。

Since IL-10 has recently been shown to exhibit pleiotropic effects on human monocytes, it was of interest to determine the effect of this cytokine on prostaglandin E2 (PGE2) production by monocytes. Recombinant IL-10 (rIL-10) did not significantly affect PGE2 production by lipopolysaccharide (LPS)-unstimulated monocytes, but efficiently inhibited PGE2 production by LPS-stimulated monocytes. The B lymphocyte–associated adaptor protein 32 kD in size (Bam32) is expressed at high levels in germinal center (GC) B cells. It has an NH2-terminal src homology 2 (SH2) domain which binds phospholipase C (PLC)γ2, and a COOH-terminal pleckstrin homology (PH) domain. Thus, Bam32 may function to integrate protein tyrosine kinase (PTK) and phosphatidylinositol 3-kinase (PI3K) signaling pathways in B cells. To further define the role Bam32 plays in B cells, we generated Bam32-deficient DT40 cells.
The B lymphocyte adaptor molecule of 32 kDa (Bam32) is an adaptor that plays an indispensable role in BCR signaling. In this study, we found that upon BCR ligation, Bam32 is recruited to the plasma membrane where it associates with BCR complexes and redistributes and internalizes with BCRs.
