연구실 소개

권용훈 교수의 연구실은 주로 유기합성화학과 생물학적 타겟을 연계한 약물 개발을 핵심으로 삼고 있습니다. 특히 코로나19 바이러스의 핵심 단백질인 Nsp15를 타겟으로 한 천연물질의 항바이러스 활성 스크리닝과, 복잡한 자연물의 전체 합성 기법을 통해 구조 규명과 입체화학 확정을 수행합니다. 또한 고도로 제어된 반응 조건을 활용한 입체선택적 합성 전략과, 불안정한 구조를 안정화시키는 전략적 보호기 기법을 응용하여, 항생제나 해양 마크로라이드 계열의 생체활성 물질을 효율적으로 합성하는 데 주력하고 있습니다.

A domino potassium permanganate-interrupted Nazarov reaction to yield syn-2,3-disubstituted 1,4-diketones via a decarbonylative cleavage of the Nazarov oxyallyl intermediate, believed to be without precedent, is presented. This process allows syn substituents to be established stereospecifically on the 2-carbon bridge connecting the ketone carbonyl carbons, and the formation of one carbon-carbon and two carbon-oxygen bonds. Two carbon-carbon bonds are cleaved in this process.

Trimethylaluminum and molecular oxygen are used in tandem to interrupt the Nazarov cyclization. α-Hydroxycyclopentanones are produced in moderate to good yield as a mixture of epimers. This sequence is the first example of combined nucleophilic/electrophilic trapping of the Nazarov oxyallyl intermediate.

We report the first total synthesis of micrococcin P2 (MP2, 1) by a diversity-oriented route that incorporates a number of refinements relative to earlier syntheses. Biological data regarding the activity of 1 against a range of human pathogens are also provided. Furthermore, we disclose a chemical property of MP2 that greatly facilitates medicinal chemistry work in the micrococcin area and describe a method to obtain MP2 by fermentation in <i>B. subtilis</i>.

Abstract Lewis acid catalyzed 4π-electrocyclization of Donor–Acceptor Stenhouse Adducts (DASAs) has been developed. By employing catalytic quantities of Dy(OTf)3, a highly efficient transformation of acyclic DASAs into the corresponding cyclized isomers was achieved.
