Professor Yongseok Choi's research lab specializes in medicinal chemistry and drug discovery, with a focus on repurposing existing drugs and developing novel small-molecule therapeutics for inflammatory and autoimmune diseases. The lab investigates epigenetic modulators, such as histone deacetylase (HDAC) inhibitors, and targets key proinflammatory cytokines like interleukin-6 (IL-6) to address unmet medical needs. A major emphasis is placed on designing and synthesizing conformationally locked nucleosides and bioactive compounds with improved pharmacological profiles, including oral bioavailability and reduced toxicity.

Abstract In recent years, the emergence of biologically active compounds that contain a heterocyclic ring has gained a great deal of attention among medicinal chemists. Among these, benzoxazole‐based compounds are particularly attractive because of their wide range of pharmacological activities. In light of the anti-inflammatory properties of histone deacetylase (HDAC) inhibitors, such as suberoylanilide hydroxamic acid (SAHA) and trichostatin A (TSA), we examined a new HDAC inhibitor KBH-A42 for its anti-inflammatory activities. KBH-A42 showed noteworthy anti-inflammatory properties in vitro via suppression of the production of TNF-alpha, a proinflammatory cytokine, and nitric oxide (NO), a proinflammatory effector molecule, in LPS-stimulated RAW264.7 cells and peritoneal macrophages.

The present work describes some recent approaches to the syntheses of three classes of locked-North nucleosides: beta-D-ribo-, beta-D-deoxyribo-, and beta-D-dideoxyribonucleosides. The method developed for the latter class permitted access to a novel bicyclo[3.1.0]hexene-type nucleosides structurally similar to D4T and carbovir. A structural analysis and biological activities are discussed.
It was examined whether hepatocyte cell lines can be used for ammonia removal in mammalian cell cultures. It was found that there exists a critical ammonium concentration level for each hepatocyte cell to remove ammonia. Among the cells tested in this work, primary hepatocytes showed the strongest ammonia removal capability if ammonium concentration is higher than the critical level.
