Professor Eunjin Ryu's research lab focuses on the molecular mechanisms governing DNA replication fidelity and genome stability, with a central emphasis on the regulation of proliferating cell nuclear antigen (PCNA) dynamics. The lab investigates how clamp loaders, particularly the ATAD5-RFC-like complex (ATAD5-RLC), control PCNA loading, unloading, and ubiquitination states to ensure accurate DNA replication and repair. Key research directions include the structural and functional regulation of ATAD5, its interactions with chromatin readers (e.g., BRD4), deububiquitinating enzymes (e.g., USP1-UAF1), and RNA helicases in resolving transcription-replication conflicts and R-loops. The lab employs single-molecule imaging, structural biology, and biochemistry to dissect the spatiotemporal control of PCNA cycling and its impact on genome integrity.

14Replication-Factor-C (RFC) and RFC-like complexes (RLCs) mediate chromatin engagement of the proliferating cell nuclear antigen (PCNA).
