Professor Sang-Hoon Han's research lab focuses on developing novel therapeutic agents targeting infectious diseases and cancer, with a strong emphasis on structural biology and drug discovery. The lab investigates antimicrobial resistance, particularly in multidrug-resistant pathogens like *Acinetobacter baumannii* and *Klebsiella pneumoniae*, and explores innovative strategies to overcome cancer drug resistance through novel microtubule inhibitors. Structural insights from X-ray crystallography are central to their rational drug design, especially in targeting viral proteases such as SARS-CoV-2 3CLpro. The lab also examines host-pathogen interactions, including viral reactivation in critical illness and the immunological responses in sepsis and immunocompromised patients.

15Starting from the MLPCN probe compound ML300, a structure-based optimization campaign was initiated against the recent severe acute respiratory syndrome coronavirus (SARS-CoV-2) main protease (3CL<sup>pro</sup>). I present the only comprehensive review of hepatic abscess from Klebsiella pneumoniae. The world literature contains 46 individually reported cases and 3 large studies of K pneumoniae liver abscess. The success of cancer chemotherapy is limited by multidrug resistance (MDR), which is mainly caused by P-glycoprotein (P-gp) overexpression. In the present study, we describe a novel microtubule inhibitor, 5-(N-methylmaleimid-3-yl)-chromone (SPC-160002), that can be used to overcome MDR. Regardless of cognitive status, altered local intrinsic activities were found in HIV patients.
