Professor Sang Hyub Lee's research lab specializes in gastrointestinal and hepatobiliary oncology, with a focus on molecular mechanisms underlying pancreatic and bladder cancers, as well as liver cystic tumors. The lab investigates tumor biomarkers, epigenetic alterations (such as NPTX2 methylation), and epithelial-mesenchymal transition (EMT) markers to improve prognosis prediction and therapeutic decision-making. Key research directions include identifying genetic and epigenetic drivers of cancer progression, evaluating chemotherapy regimens in metastatic pancreatic cancer, and refining diagnostic criteria for complex biliary and pancreatic lesions.

15Fas (Apo-1/CD95) is a cell-surface receptor involved in cell death signaling. The key role of the Fas system in negative growth regulation has been studied mostly within the immune system, and somatic mutations of Fas in cancer patients have been described solely in lymphoid-lineage malignancies. We analyzed somatic mutations and loss of heterozygosity of Fas gene in 43 transitional cell carcinomas of urinary bladder. The expression of markers of EMT and metastasis in PDACs was significantly associated with pathologic features of aggressiveness. CD24 and S100A4 expressions were significant predictors of poor survival; thus, immunohistochemistry for these markers in resected specimens may help to identify PDAC patients with a poor prognosis.
In hepatic cysts with mural nodule, left-lobe cyst or increment of serum alkaline phosphatase seem to be indicative of BCTs in the diagnosis of suspicious hepatic cyst. Intracystic debris, bile duct dilation, and mural nodule may be suggestive clinical features of malignancy in BCTs.
The aberrantly methylated NPTX2 gene may help to distinguish between chronic pancreatitis and pancreatic cancer with conventional diagnostic tools and could become a valuable diagnostic marker.
An impacted stone, stone size (>or=30 mm) and stone size to bile duct diameter ratio (>1.0) were predictors of failure of endoscopic mechanical lithotripsy for a difficult bile duct stone. Alternative approaches should be considered in patients with predictors of unsuccessful endoscopic mechanical lithotripsy in order to avoid wasting time and resources.
