Professor Soong Deok Lee's research lab specializes in molecular forensic genetics and epigenetics, focusing on the analysis of degraded and low-copy number DNA from challenging sources such as ancient and postmortem skeletal remains. The lab develops advanced molecular techniques for STR typing and DNA methylation profiling to improve the identification of individuals, including monozygotic twins and disaster victims. Key research directions include optimizing DNA extraction and amplification protocols, applying massively parallel sequencing for degraded DNA, and investigating mitochondrial DNA alterations in cancer. We investigated whether a combination of recently introduced methods, total demineralization and ion-exchange columns, would increase DNA recovery from old bone. Ten bone samples taken after a burial period of approximately 60 years were used in this study. Bone powder was digested using total or incomplete demineralization. The effects on STR typing results, using a two-step PCR protocol that combines the annealing and extension steps, were determined using samples at various DNA concentrations. BACKGROUND: Short tandem repeat (STR) markers cannot be used to distinguish between genetically identical monozygotic (MZ) twins, causing problems in a case with an MZ twin as a suspect. Many studies have shown that in older MZ twins, there are significant differences in overall content and genomic distribution of methylation.
